Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Cytokeratin 13 (KRT13) is the major acidic keratin, which together with KRT4, its basic partner, is expressed in the suprabasal layers of non-cornified stratified epithelia including tongue mucosa, esophagus, anal canal epithelium, tracheal epithelium, uterine cervix, and urothelium. Defects in the KRT13 gene are a cause of white sponge nevus of cannon (WSN), a rare autosomal dominant disorder, which predominantly affects non-cornified stratified squamous epithelia and is characterized by the presence of soft, white and spongy plaques in the oral mucosa. KRT13 has been used as a marker for non-keratinized squamous epithelium. It is also expressed in various squamous metaplasia, but it is down regulated in squamous dysplasia and squamous carcinoma. This antibody recognizes a protein of 40 kDa, identified as Thyroid transcription factor-1 (TTF-1). TTF-1 is a member of the NKx2 family of homeodomain transcription factors. It is expressed in epithelial cells of the thyroid gland and the lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testes, pituitary, prostate, and adrenal glands are unreactive. Anti-TTF-1 is useful in differentiating primary adenocarcinoma of the lung from metastatic carcinomas originating in the breast, mediastinal germ cell tumors, and malignant mesothelioma. It can also be used to differentiate small cell lung carcinoma from lymphoid infiltrates. Loss of TTF-1 expression in non-small cell lung carcinoma has been associated with aggressive behavior of such neoplasms. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. This antibody recognizes a single chain glycoprotein of 105/120 kDa, identified as CD30/Ki-1. CD30 is synthesized as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkin's disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This MAb distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas.
